Service description
This service includes the following steps:
A/ Evaluation of vector* activity
1. Microinjection of the construct* into one-cell stage embryos.
2. Evaluation of vector activity in 24-48hpf embryos with a fluorescence scope.
Delivery time: 2 weeks.
Delivery format: The customer will receive photos from F0 embryos at 24-48hpf to evaluate the fluorescence activity of the provided vector. Approval by the customer is required in order to proceed with the following step.
* The Functional Genomic Service does not provide cloning services. Contact the manager for advice on vector selection and/or cloning.
The construct is to be provided by the customer. We require 20µl at a minimum concentration = 50ng/µl (from kit).
Note: Without obvious technical issues, the price for step A (see below) will be charged independently of the final outcome.
B/ Generation of a zebrafish or medaka stable transgenic line
1. Raising of F0 positive embryos to adulthood.
2. Screening of F0 adult fishes:
- Outcross: individual F0 males and females are outcrossed with WT females and males respectively to identify Founders.
- Selection of fluorescence positive F1 embryos from founder/s.
Delivery time: approximately 6 months.
Delivery format**: The customer will receive bleached F1 transgenic embryos.
