Staining in paraffin sections allows detailed analysis of tissue morphology and composition. Hematoxylin-eosin (H&E) is the most widely used in general histology; hematoxylin stains cell nuclei blue/purple, while eosin highlights the cytoplasm and extracellular fibers in pink tones. Nissl staining is used in nervous tissue, highlighting Nissl bodies (rough endoplasmic reticulum) in neurons, aiding in neuronal integrity studies. PAS (Periodic Acid-Schiff) stain marks carbohydrates, glycogen, and mucins, making it useful for epithelial, kidney, liver, and intestinal tissues, as well as basement membrane identification. Masson¿s trichrome differentiates collagen (blue or green) from muscle and cytoplasm (red), allowing fibrosis and connective tissue studies. Sirius Red stain is specific for collagen fibers, enabling their analysis through polarized light microscopy, making it useful in fibrosis-related diseases. Van Gieson stain highlights collagen in red and muscle in yellow, helping evaluate extracellular matrix changes and fibrosis in cardiac and connective tissues. These techniques are essential for pathological diagnosis, biomedical research, and studying structural alterations in various organs.
