PIF2024 - MOLECULAR AND PHARMACOLOGICAL REGULATION OF VIRAL EPISOMAL DNA EXPRESSION (PID2023-148194OB-I00)

Episomal DNAs are found in the nucleus as consequence of virus infections and chromosome disruptions, and they are in many cases associated to the development of disease, including cancer, or are being used as vectors in viral gene therapy strategies to treat genetic disorders. Selective regulation of episome-derived gene expression has been recently proposed as a novel approach for episome-related disease treatment. In this context, we have recently identified cellular Senataxin protein as key negative regulator or episome-derived gene expression. Here, we propose to deepen in the understanding of the mechanisms regulating episomal expression in hepatitis B virus and adeno-associated virus episomes. We will combine deterministic and unbiased approaches to discover new molecular targets and small molecules for drug development. The trainee will have the opportunity to contribute in experimental, technical and intellectual aspects of the project. S/he will enroll the Molecular Biosciences doctoral program at UAM and will participate in the CNB Thesis Advisory Committee program. This program is part of the PhD Researchers Career Development Plan which provides with specific training in scientific integrity and career development. In addition, the student will participate in weekly labmeetings and journal clubs, departmental seminars and other CNB-organized seminars given by national and international experts in their respective fields to which students are encouraged to attend.
